Important Information for Prospective CElixir Users
CElixir family of products has been optimized to perform best when used with positively charged or amphoteric
negatively charged, neutral compounds or mixtures of all.
Although most parameters affecting CElixir CZE will be the same as when used with Bare Fused Silica; CZE with
CElixir is different. It is important that you NOT simply substitute CElixir into your current method
for your run buffer. To get the most out of CElixir’s features, you should develop a new method and follow our
method development steps listed below. You should find the pH at which your sample’s separation is best with
CElixir.
- Following the instructions in the CElixir Operating and Trouble Shooting Manual, select the CElixir
Accelerator Solution (B) that is two points lower than your analyte’s pK or pI. If you do not know the pK or pI,
start with Accelerator Solution (B) pH 2.5. Include in this run, a neutral marker to determine EOF along with the
migration time of your peaks.
- Run the same method as above but this time dissolve your sample in the next higher pH of CElixir
Accelerator Solution (B). Note EOF and migration time of your peaks.
- Repeat the above step with each available Accelerator Solution (B) until you operate with CElixir at
pH 9.2.
- Find two runs where you achieved your best resolution and efficiency. It is most likely that your optimized run
is using CElixir at the pH somewhere between these two runs.
- Follow the instructions on page 24 of the CElixir Operating and Trouble
Shooting Manual to adjust the Accelerator Solution (B). Continue to bracket the pK or suspected pK until you are
satisfied with the results.
